KPV (Lysine–Proline–Valine) is a naturally occurring tripeptide fragment derived from the C-terminal region of α-melanocyte-stimulating hormone (α-MSH). It consists of three amino acids—lysine, proline, and valine—and retains significant anti-inflammatory activity independent of the parent hormone’s melanocortin receptor signaling. Mechanistically, KPV has been shown in preclinical models to reduce inflammation by inhibiting NF-κB activation, decreasing pro-inflammatory cytokines such as TNF-α, IL-1β, and IL-6, and modulating immune cell activity. Unlike full-length α-MSH, KPV does not significantly stimulate melanogenesis, making it a targeted anti-inflammatory fragment. It has been investigated primarily in experimental models of inflammatory bowel disease, dermatitis, and other inflammatory conditions due to its ability to downregulate inflammatory signaling pathways while demonstrating a favorable safety profile in early research contexts.
